Stem cell therapy has demonstrated a great future for the disease treatment. However,the delivery of stem cells to targeted tissues is still one of the key difficulties.The injection and surgical transplantation are currently mainly used delivery strategy in stem cell therapy.?As known, epidermal stem cells play a centrol role in homeostasis,wound repair and gene therapy.It resided in the basal layer of epidermis.In addition, nearlly all of the skin cancers begin in the epidermis of the skin. But so far, there is no an ideal strategy to deliver cells for such targeted rigions. Laser microporation is a novel method for transdermal delivery of macromolecules. Recently, we reported the transdermal delivery of antibodies using this technology for the first time. The aim of this study is to deliver stem cells to the epidermis using laser microporation technology and to discover the effects of transdermal delivery of stem cells in vitro and in vivo. This study could provide a noval strategy for stem cell delivery and improve the efficiency and application of stem cells to treat skin cancer, refractory single gene genetic skin diseases and skin damage repair treatment.
