心房颤动(房颤)是最常见的心律失常之一。心房重构是房颤复发和维持的重要机制,炎症和氧化应激在房颤的发生和维持中发挥重要作用。罗格列酮具有抗炎抗氧化作用,可能改善心房电重构和结构性重构,预防房颤的发生。本课题拟建立四氧嘧啶介导的兔糖尿病模型,应用离体电生理、膜片钳、定量PCR、Western-blotting等实验方法,从整体水平、细胞水平和离子通道水平及分子生物学水平评价罗格列酮对糖尿病兔心房结构性重构和电重构的干预作用及其可能机制,同时探讨炎症和氧化应激、心房肌纤维化、离子通道电流变化和蛋白表达变化在糖尿病导致房颤中的地位和作用,为房颤的防治提供新思路。罗格列酮对糖尿病心房重构的干预作用国内外未见报道
