Description
Heart failure is associated with changes in the surface expression and regulation of the Calcium
channel CaV1.2, which contributes to the exertional limitations and propensity for arrhythmia
experienced by patients with this disease. Combining APEX proximity labeling with quantitative TMT
SPS MS3, I will map the changing “neighborhood” of CaV1.2 in chronic heart failure. Using genetic
models of APEX-defined candidate proteins, cellular electrophysiology, and an innovative transgenic
mouse model engineered to assess CaV1.2 surface expression, I will gain deep insights into the
molecular and cellular pathophysiology of heart failure.
